SHINVA Bioreactor
  1. General
This SHINVA bioreactor, developed by BIO-YD, is used for effective cell culture in pharmaceutical industry and meets requirements of medical & health and cGMP. It is a very ideal for various cell cultures with its functions, reliability, convenience and economy.  
 
Advantages and features of SHINVA Bioreactor:
u Tank body is self-designed by BIO-YD  and manufactured according to standards of ASME BPE and GB150 with inner polishing(Ra<0.4µm) and outside sandblasting treatment. Pipeline reliably and clearly arranged to meet 3D requirement and effectively avoid dead leg. This whole system is modularly designed and installed with space saving.
u The stirring system applies magnetic stirring unit, a patented product of BIO-YD. Static sealing is used in stirring with its excellent tightness and zero leakage to make bioreactor totally isolated with the outside, ensuring safety with long-term sterile operation and free maintenance and easy cleanness ; meeting much more requirements of pharmaceutical industry.
u DO in deep aeration is achieved by micro-bubble aeration device and bubble-free aeration device. Aeration mode is optional. Micro-bubble aeration mode makes micro and fair bubbles. Bubble-free aeration mode makes no bubble and no shear force to cell, which meet aeration requirement in cell growth.
u Cell trapping unit, which is equipped with rotary filter and tank-bottom-valve trapping piece, achieves continuous harvest of virus solution in continuous culture. Cell trapping unit can effectively separate virus solution with micro-carrier and cell to achieve harvest of virus solution. Tank-bottom-valve is specially designed with two outlets. The rest solution after tank top harvest is harvested by this tank-bottom-valve trapping piece to furthest improve product yield.
u It is easy to use peristaltic pump and tank strip sight glass that with level display to control liquid volume and then to achieve perfusion.
u SIP is achieved by steam with high level automation. It is also easy to sterile manually.
u Temperature control system applies plate heat exchanger and electric heater to make sure sensitive, stable and convenient control.
u Critical facilities like electrode, instruments, controller and valves and elements are imported, guaranteeing bioreactor reliability and stability.
u Touch screen makes parameters display, control, calibration, storage, inquiry realizable. It is functional and easy to use.
 
2.     Function  Specification
SHINVA bioreactor system includes three parts: tank body, pipelines and control cabinet. Applying PLC software achieves control to stirring, temperature, PH, DO, antifoam, tank pressure and SIP, meanwhile data record and inquiry function.
  1. Bioreactor
SHINVA vertical 3-layer tank (shell/jacket/isolation layer) .SS316L is for shell, SUS 304 is for jacket and isolation layer. There are four baffles, top sight glass, lamp glass, stirring hole and ports in/on tank.
  1. Cleaning Control
Cleaning spare port connected with external solution (lye, WFI). Run cleaning in according to specified program. Cleaning objects are feeding pipelines, outlet pipelines, inlet pipelines (off-line cleaning for filter), exhaust pipelines (off-line cleaning for filter), inner tank.
  1. Sterilization Control
SIP device connected with external dry PW and industrial steam for on-line SIP. SIP objects are feeding pipelines, outlet pipelines, sampling pipelines, inlet pipelines (filter included), exhaust pipelines (filter included), inner tank. SIP temperature: ≥121℃, time of SIP : 30min. (settable).
  1. Feeding Control
There are six feeding pipelines: lye feeding, alkali feeding, antifoam feeding, inoculation/syrup feeding, and medium/solution feeding and special supplies. All feedings are provided by peristaltic pump with manually adjustable flow. Feedings are sterile.
  1. Gas Inlet Control
Four gases inlet, they are compressed air, oxygen, CO2 and N2. Gas pipelines are equipped with gas relief valve, filter, pneumatic diaphragm valve, solenoid valve, mass flow controller and rotary flow meter. Gas inlet is controlled by PID control mode. Micro-bubble distributor and bubble-free device are equipped for deep aeration and DO.
  1. Exhaust Control
Condensation section, filter with house heater, pneumatic diaphragm proportional valve are equipped to make sure no gas pollution to the environment.
  1. Stirring Control
Upper magnetic stirring system is applied with servo motor and driver. Variable frequency with stirring speed 0-400rpm. Low shear force blades are used to reduce damage to cells.
  1. Temperature Control
Jacket is introduced to do temperature control. Temperature control device is equipped with circulation pump, plate heat exchanger and electric heater whose grouping control improves sensitivity and stability of temperature control. Temperature control of bioreactor is based on double control of tank temperature and jacket temperature to make stability. Accuracy of temperature control: ±0.2℃.
  1. pH Control
 PID control is applied to achieve automatic pH control. PH control is connected with CO2 control valve and acid/alkali peristaltic pump. Flow of CO2 is adjusted by flow controller, and flow of acid/alkali is adjusted by PLC to further improve sensitivity of pH control.
  1. DO Control
   PID control is applied to achieve automatic DO control. DO control is connected with O2 flow controller, CA flow controller and stirring unit. By adjusting opening of flow controller, automatic DO control is achieved. O2 flow and CA flow can be adjusted by flow controller and solenoid valve to further improve sensitivity of DO control.
  1. Antifoam Control
Tank foam is controlled by correlation of antifoam level and deformer peristaltic pump.
  1. Pressure Control
Controller of inlet flow and automatic proportional valve of exhaust are applied to control inner tank pressure. PID control mode achieves automatic control of tank pressure and in-tank constant flow ventilation by controlling the opening of flow controller and on-off rate of regulating valve.
  1.  Sampling
Sampling device is used for multiple and sterile sampling.
  1.  Liquid Transferring
    Liquid transferring device that is equipped with cell trapping system is used for taking liquid form tank top or draining form tank bottom. Liquid transferred by compressed air, and constant pressure maintains after reached the set value.
  1.  Waste Drainage
Condensate drainage pipeline and waste water drainage pipeline are separately installed.
  1. Control System
Control functions of bioreactor is meet by control cabinet, which including PLC controller, display screen, DO controller, pH controller and data recording/storage etc..
 
  1. Display Screen
LCD touch screen for basic display and control of system parameters.
  1. User Permission
Three-level user permission, with passwords protection.
  1. Flow Chart Display
Operation flow chart of bioreactor shows parameters, valves and operation condition. Parameter setting and operation status can be manually modified.
  1. Control Display
Display operation parameter and modify parameter setting.
  1. Operation Program Display
Display cleaning program, empty sterilization program, feeding program, real sterilization program, temperature-control program of jacket, gas inlet program, and material transferring program. All programs are automatically controlled.
  1.  PH Controller
Independent pH controller is used for temperature compensation, pH value display and display/modification of pH setting value.
  1. DO Controller
Independent DO controller is used for temperature compensation, DO value display and display/modification of pH setting value.
