Transfection Protocol
 

  1. Description
 
CHR (see note A) is a revolutionary transfection reagent that isn't made from cytotoxic PEI. CHR has been developed to cater for many cell lines. Arginine residues play a critical role in intracellular uptake. Histidine has buffering capacity in the acidic environment of endosomes. Disulfide bonds were reported to show significantly increased transfection efficiency and cell viability.
 
  1. Cell Lines Successfully Transfected Using CHR:

 Ina dessicator dryer with anhydrous calcium chloride at RT
 
  1. Recommended Protocol
 
We recommend using a **-well plate format to optimize transfection conditions for a particular cell type.
 
Plate cells one day before the transfection experiment so that cells will be approximately *5% confluent on the day of transfection.
 
Preparation of CHR& pDNA complexes:
CHR solution: Combine *0 mg CHR with *0 ml DMEM (pH 6.5 for CHO, see note B).
Combine 0.5 ug pDNA with *0 uL DMEM.
*0 uL CHR solution was added to this *0 uL pDNA-DMEM gently and incubated for *0 minutes at room temperature.

*0 uL CHR /pDNA complexes were added into each well while gently swirling the plate.
Incubate cell at *7 C in a CO2 incubator. The transfection efficiency of reporter gene could be analysis at ****8 hours after adding the complexes.
 
Gene transfection in suspension cells:
At 1hour after cell seeding, the CHR/pDNA complexes can be added into cells followed transfection assay at ****8 hours after transfection.
 
Note A
CHR free base cannotget any transfection, only CHR HCl can transfect cells.
