Camptothecin is an alkaloid isolated from the stem wood of the Chinese tree, Camptotheca acuminata. This compound selectively inhibits the nuclear enzyme DNA topoisomerase, type I. Several semisynthetic analogs of camptothecin have demonstrated antitumor activity.

Camptothecinis a naturally occurring alkaloid derived from the plant Camptotheca acuminata that was identified in an anticancer drug discovery screen in the ***0s. Camptothecin forms a stable ternary complex, preventing normal DNA re-ligation, and causes the complex to collide with the replication fork, leading to break and cytotoxicity.
Camptothecins are mainly used in lung, breast, and brain cancer. However, topotecan is approved by the USFDA for the treatment of lung and ovarian cancer and irinotecan is approved for the treatment of colon cancer.
